Tianeptine Sodium and the extreme difference in potency
« on: January 10, 2019, 12:28:33 PM »
Hey everyone,

Having a lot of experience with this chemical I have noticed something peculiar. The potency varies incredibly vendor to vendor. What is strange about this is that my company's independent testing shows very little difference in purity. All batches I have received have tested at least >98.9% and usually greater than 99.7%. I became determined to figure out this mystery and I have come up with what I feel, is the explanation.

Tianeptine Free Acid is very susceptible to trans esterfication. The Sodium is always made from the free acid. If the chemist is using either methanol or ethanol during synthesis while the solution is either basic or acidic trans esterfication will occur. It is well known that the esters of acids are almost always much more pharmacologically active. I suspect that the very strongly opioid producing batches have higher levels of these esters.

An example is Demerol which is an Ester. The acid of Meperidine is very weak or even inactive. Another example is Ritalinic Acid compared to methylphenidate.

Anyone have any insight?
